This is a timely and well presented, for the most part, review on a cutting-edge topic. I have no major criticisms. However, I have a few suggestions for changes in the report. - First, all over the manuscript, the authors should write the full name of all abbreviations in their first mention in the text (even if most are written at the end of the manuscript). As a review article, it should be enough for future readers to catch all the related data in one area and avoid distraction related to searching about what do the mentioned abbreviation(s) stand for? Just for example: -p.3; line 100: HOTAIR, line 102: H3K4me2, GATA-AS, line 104: LOX12, line 111: SOX18, SMAD6, COUP-TFII,line 115: SINE, line 119: GAS%, line 125: HUVECs, Line 133: SINEB2, line 135: BACE1AS,line 141: sONE.........etc. Please revise each page meticulously as not all future readers should have related specialty. - After spelling out all the abbreviations, the authors should confirm that all are included in the abbreviation list at the end of the manuscript. - Some sentences require revision like (p.2; line 48” - The authors should cite more than one reference in the following text: • p. 2; line 79: “Despite there were controversial findings reported in previous lncRNAs studies (14)”, and • p.2; line 89: “numerous studies revealed that their cellular localization is the primary determinant of their functions (17)” • p. 3; line 96: “Unfortunately, only a few identified lncRNAs were functionally characterized by previously published studies” - Page 2; line 83: please add (pol II) after “RNA polymerase-II”, to facilitate the following of the authors in the subsequent sections. - Page 3; line 117: please replace the word “up-expressed” by “up-regulated” or “over-expressed” - Page 3; line 141: add “s” to “endothelial gene”. - Each table should be self-exploratory; all mentioned abbreviations included in the table should be written in detail in the foot table. In addition, for the future reader, the authors should refer to in the figure legends that all abbreviations mentioned in the figure are spelled out at the end of the manuscript. - Tables 2 and 3 are preferable to be positioned after their specified text to facilitate following the authors in their elaboration. - p.8: line 255: revise “although RNAi has been shown to be effective in the nuclease”. - p. 9; line 327: the authors should revise the citation of “Carlevaro‑Fita, J., & Johnson, R.”. The initials of the authors are not written in the text. - p. 10; line 336: add the word “fluorescence” to the long name of “smRNA‑FISH” - p.11; line 356” what is meant by “RN‑enrich”? - p.11; line 369: please correct “signal‑to‑nose ratio”. - p. 11; line 370 Is the following statement “For the first time, ChIRP method was used to identified Xist binding site on X‑chromosome (160)”, should be positioned her or in the previous section (4.3.2.2.)?
