Content of review 1, reviewed on August 26, 2014

The database presented by Pellegrini and co-workers is related to a comprehensive sequencing of the black truffle T. melanosporus. The experimental details concerning the preparation of the libraries are well reported. There is not much about the biological effects of the extensive methilation and transposons on general gene expression and physiology, but I guess that these scientific questions will be addressed in a supplementary work.

Minor revisions:

Pag. 3. A. immersus should be written in a complete form (Ascobolus immersus?). There are different double spaces in the text. Please check it.

Discretionary Revisions:

The genome of T. melanosporus is quite big, but it expresses “only” 7500 putative genes. Using your RNA-seq data it is possible to detect additional transcripts that were not previously annotated?

Level of interest:

An article whose findings are important to those with closely related research interests Quality of written English Acceptable Statistical review No, the manuscript does not need to be seen by a statistician. Declaration of competing interests I declare that I have no competing interests' below.
